Working Principle
(a). In HPLC, the pump is responsible for delivering the mobile phase at a controlled flow rate.
(b). However, air bubbles can easily dissolve in the mobile phase under standard atmospheric pressure, which can cause issues like flow rate fluctuations and baseline noise/drift. To prevent this, a degassing unit is used to remove air bubbles from the mobile phase .
(c). Once the air bubbles have been removed, the mobile phase is delivered to the column. The sample injector then introduces a standard or sample solution into the mobile phase.
(d). To ensure consistent separation of compounds in the column, a column oven is used to maintain a constant temperature.
(e). As compounds elute from the column, they are detected by a detector placed downstream of the column.
(f). The detector signal is processed by a workstation to obtain a chromatogram, which is used to identify and quantify the individual compounds.
